S-nitrosylation of SlAPX Is Involved in Alleviating Oxidative Damage in Transgenic Tobacco under Nitrate Stress

چکیده

Nitric oxide (NO) modulates plant response by post-translationally modifying proteins, mainly through S-nitrosylation. Ascorbate peroxidase (APX) in the ascorbate-glutathione (AsA-GSH) cycle participates removal of hydrogen peroxide (H2O2). However, relationship between S-nitrosylation and role tomato APX (SlAPX) under nitrate stress is still unclear. In this study, enzyme activity, mRNA expression, level SlAPX were significantly increased roots after treatment. protein could be S-nitrosylated S-nitrosoglutathione vitro, activity was The overexpressed tobacco plants grew better than wild type (WT) stress. Meanwhile, transgenic showed lower reactive oxygen species malondialdehyde content, higher APX, monodehydroascorbate reductase, glutathione reductase activities, ascorbic acid/dehydroascorbic acid, reduced glutathione/oxidized ratio, proline, soluble sugar contents those WT Moreover, seeds tolerance to methyl viologen induced oxidative compared with WT. NO accumulation Our results provide novel insights into mechanism modulation excess involving modification.

Protein S-nitrosylation in plants under abiotic stress: an overview

Abiotic stress is one of the main problems affecting agricultural losses, and understanding the mechanisms behind plant tolerance and stress response will help us to develop new means of strengthening fruitful agronomy. The mechanisms of plant stress response are complex. Data obtained by experimental procedures are sometimes contradictory, depending on the species, strength, and timing applied...

Dehydrins Impart Protection against Oxidative Stress in Transgenic Tobacco Plants

Environmental stresses generate reactive oxygen species (ROS) which might be detrimental to the plants when produced in an uncontrolled way. However, the plants ameliorate such stresses by synthesizing antioxidants and enzymes responsible for the dismutation of ROS. Additionally, the dehydrins were also able to protect the inactivation of the enzyme lactate dehydrogenase against hydroxyl radica...

Protein Expression Regulation under Oxidative Stress*□S

Oxidative stress is known to affect both translation and protein turnover, but very few large scale studies describe protein expression under stress. We measure protein concentrations in Saccharomyces cerevisiae over the course of 2 h in response to a mild oxidative stress induced by diamide, providing detailed time-resolved information for 815 proteins, with additional data for another 1,100 p...

High glucose attenuates protein S-nitrosylation in endothelial cells: role of oxidative stress.

OBJECTIVE Hyperglycemia-induced endothelial dysfunction, via a defect of nitric oxide (NO) bioactivity and overproduction of superoxide, is regarded as one of the most significant events contributing to the vascular lesions associated with diabetes. However, the mechanisms underlying such hyperglycemic injury remain undefined. We hypothesized that alterations in cellular protein S-nitrosylation...

S-nitrosylation of peroxiredoxin 2 promotes oxidative stress-induced neuronal cell death in Parkinson's disease.

Peroxiredoxins (Prx), a family of peroxidases that reduce intracellular peroxides with the thioredoxin system as the electron donor, are highly expressed in various cellular compartments. Among the antioxidant Prx enzymes, Prx2 is the most abundant in mammalian neurons, making it a prime candidate to defend against oxidative stress. Here we report that Prx2 is S-nitrosylated (forming SNO-Prx2) ...
